My nominees:
"When Doves Cry" by Prince
"Lucky Star" by Madonna
"Like a Virgin" by Madonna
"Like a Prayer" by Madonna
"Billie Jean" by Michael Jackson
"Jump" by Van Halen
Any other nominees, with reasons as to why?

'Where's My Mind' by the Pixies - the sound of the birth of Grunge
'F*ck the Police' - N.W.A. - the sound of Gangster rap being born
Those were probably the big genre-defining songs of the decade, both being released in the late 80s.
The early and mid 80s had many important singles, but perhaps not as influential as these two.
